Protease Research Our custom services are provided to companies and
individual researchers on a contract or fee-for-service basis. We

We offer a complete selection of peptide substrates for your protease. We use phage display technology to screen an in-house library of 1.8x108 independent clones (Cloutier et al. 2002. Eur J Biochem 269:2747-54) to determine pentapeptide sequences specific for your protease of choice. Synthesis of specific fluorescent protease substrates We construct for you protease substrates, which can be used in Med Discovery's RFPS system (Felber et al. 2004. Biotechniques 36(5):878-85). Protease substrate sequences might be customer designed or selected by Med Discovery using phage display. Determination of enzymatic activities by fluorescent assays We use our in-house RFPS system to calculate kinetic parameters (kcat/km) of peptidic substrates towards proteases of your choice. Development of specific inhibitors We showed that specific modification of the reactive site loop of
natural protease inhibitors can create novel protease inhibitors
with high specificity and activity (Cloutier et al. 2004. Eur J Biochem
271:607-13). Using phage display selected or customer defined peptide
substrate sequences specific for the protease of choice we engineer
chimeric protease inhibitors based on natural proteins. |
